A japa mala has 108 beads and one larger bead, the meru or sumeru, which marks the beginning and end of a round.
Hold the mala in the right hand and move one bead per repetition. When you reach the meru, traditionally you turn the mala rather than crossing over it.
Consistency matters more than number: a short, attentive practice each day is the usual advice.
